Abondance et composition du phytoplancton sur les lagunes méditerranéennes oligo et mésohalines ArchiMer
Derolez, Valerie; Bec, Béatrice; Grillas, Patrice; Sanchez, Ana Elena; Raimbault, Patrick.
In the context of WFD implementation on Mediterranean lagoons, work has been carried out since 2010 by Ifremer, Tour du Valat and IRSTEA (Onema and AERM & C fundings), to assess the relevance of macrophytes and macrofauna indicators for oligohaline and mesohaline lagoons (mean salinity <18 PSU). The question of the relevance of indicator metrics and thresholds for the "phytoplankton" biological quality element also arises, especially due to the confined nature of olgo-mesohaline lagoons. The objective of this study is to characterize the abundance and composition of phytoplankton on 5 oligo and mesohaline lagoons of the French Mediterranean coast (Scamandre, Crey, Charnier, Bolmon and Grand Palun). Analyses of accessory pigments (chlorophyll and...

Impact of diuron on aneuploidy and hemocyte parameters in Pacific oyster, Crassostrea gigas ArchiMer
Bouilly, Karine; Bonnard, Marc; Gagnaire, Beatrice; Renault, Tristan; Lapegue, Sylvie.
Diuron is a substituted urea herbicide used for agricultural and nonagricultural weed control. Its widespread use and relatively slow breakdown led us to analyze its influence on aneuploidy level (lowered chromosome number in a percentage of somatic cells) and hemocyte parameters in Pacific oysters, Crassostrea gigas. Adult oysters were subjected to two diuron concentrations (300 ng L-1 and 3 mu g L-1) for 11 weeks. Significantly higher aneuploidy level was observed in diuron-treated oysters compared with the control. Furthermore, the observed impact on aneuploidy persisted to the next generation as offspring exhibited significantly higher aneuploidy levels when their parents had been exposed to diuron. Significant increases in hemocyte parameters (cell...

Le système immunitaire du bar Dicentrarchus labrax. Immunoglobulines, cellules productrices d'immunoglobulines, ontogenèse applications à une pathologie virale néonatale ArchiMer
Breuil, Gilles.
The sea bass, Dicentrarchus labrax, has been subjected to intensive aquaculture on the Mediterranean cost for ten years. Marine fish hatcheries are key factors in the aquaculture field, and face different pathologies, especially one of viral origin (nodavirus) that could challenge their feasibility. Consequently, we have tried to provide some data on larvae and spawning stocks. Firstly, we have studied the immunoglobulins, components of the specific humoral immunity, and their role as antibodies. On adult basses, these immunoglobulins are tetrameric molecules with high molecular weight (850 kD), made of heavy (H) - 80 kD and light (L) - 26 kD chains. In the eggs, simplest forms such as monomeric or dimeric are brought to the fore; they are located under...

Study of interactions between flat oyster, Ostrea edulis, and the parasite Bonamia ostreae using flow cytometry and suppression subtractrive hybridization (SSH) ArchiMer
Morga, Benjamin; Arzul, Isabelle; Faury, Nicole; Gagnaire, Beatrice; Chollet, Bruno; Renault, Tristan.
Bonamiosis due to the intrahaemocytic protistan parasite Bonamia ostreae is a European endemic disease affecting flat oysters Ostrea edulis. Control of bonamiosis requires a good knowledge of the parasite life cycle including host-parasite interactions. In this study, we investigated interactions between parasites and haemocytes by flow cytometry and Suppression Subtractive Hybridization (SSH). In both approaches, analyses were performed after 2 hours of contact between haemocytes and parasites purified from highly infected flat oysters. Flow cytometry analyses consisted in testing haemocyte activities including esterase activities, reactive oxygen species (ROS) production and phagocytosis after contact with live parasites and parasites inactivated by...
